Mechanism of Action
Tesamorelin is a stable analog of GHRH (growth hormone-releasing hormone) with an N-terminal modification that resists degradation by the DPP4 enzyme:
1. Binds to the pituitary GHRH receptor, stimulating pulsatile release of endogenous GH; it is not direct exogenous GH
2. GH promotes IGF-1 secretion by the liver
3. Targets visceral adipose tissue (VAT) breakdown, prioritizing the reduction of deep abdominal fat; subcutaneous fat is minimally affected, resulting in minimal weight change
4. Preserves the body’s natural GH negative feedback regulation; unlike DAC-modified CJC-1295, it does not sustain elevated GH levels
