Target and Mechanism of Action
Target: GnRH receptor (GnRHR / LHRHR), a GPCR receptor in pituitary gonadotroph cells
1. Pulsed administration: Mimics endogenous GnRH pulses from the hypothalamus, stimulating the pituitary to release LH (luteinizing hormone) and FSH (follicle-stimulating hormone), thereby maintaining the hypothalamic-pituitary-gonadal (HPG) axis – pituitary – gonadal axis, thereby maintaining testosterone/estrogen levels, spermatogenesis, and follicular development
2. Continuous administration: Sustained stimulation downregulates GnRH receptors, which in turn inhibits LH/FSH secretion (the mechanism of long-acting GnRH agonists, such as leuprolide)

## Pharmacokinetic Properties
– IV half-life: 2–4 minutes; subcutaneous administration results in a very short half-life, as the drug is rapidly degraded by peptidases in vivo
– Administration rationale: Pulsed dosing is required to maintain HPG axis activity; continuous administration leads to receptor desensitization and gonadal suppression
